The Member for Bathurst replaces John Barilaro, who on Monday announced his resignation.
Nationals MPs held a vote at NSW Parliament where Mr Toole stood against Water Minister Melinda Pavey.
Mr Toole won the ballot 15-3.
Bronnie Taylor will be National deputy leader.
Nationals Whip Steph Cooke has confirmed the appointments.
"I'm really pleased to be elected as the leader of the NSW Nationals this morning, I'll have more to say very shortly," Mr Toole said after the party room meeting this morning.
"But I'll tell you what, I'm looking forward to getting back to work."
Before entering Parliament in 2011, Mr Toole worked as a primary school teacher for almost 20 years.
He was elected to Bathurst Regional Council in 2005 and became Mayor two years later.
The new leaders' first task will be the reopening of NSW, with COVID-19 restrictions set to ease on October 11.
"I think it's important that we focus on people, that we focus on communities, and we make sure that we support businesses re-opening their doors," Mr Toole said this morning.
